Characterization of Calcium Current in Smooth Muscle Cell of Rabbit Colon

Abstract
Whole cell patch-clamp technique was used to investigate the characteristics of Ca2+ currents in the enzymatically isolated colonic smooth muscle cells of rabbit. Isolated cell contracted reversibly in response to high K+ (150 mM). The
current-voltage
relationship of Ca2+ current presented the bell shape. The amplitude of Ca2+ current was dependent to extracellular Ca2+ concentration up to 5 mM. The voltage of half inactivation(V1/2) of the peak component obtained from the curve fit was -39.5
mV
and
it was suggested a presence of window current. Inactivation of Ca2+ current could be well fitted by single exponential decay. Various inorganic Ca2+ channel blockers suppressed Ca2+ current and their potencies shows the following order ;
Cb¡µCo¡µNi¡µMn.
Nifedipine (1¥ìM) decreased Ca2+ current, whereas BAY K 8644 (1¥ìM) increased Ca2+ current. The Ba2+ was more permeable than Ca2+ through Ca2+ channel.
These results suggest that charge carrier of inward current is Ca2+ through L type Ca2+ channels in smooth muscle cell of rabbit colon.
